I know the NPSL teams qualify by playing in regional USASA qualifying tournaments, but how do PDL teams qualify?
They use a mini-table from the first few matches (I think it's 5) during their league schedule. The team at the the top of the table qualifies.
Few clubs in the Lamar Hunt/U.S. Open Cup have the pedigree of the Brooklyn Italians. After all, they are two-time winners of the event, in 1979 and 1991. But head coach Joe Barone is quick to point out that this is not the Italians of the old days, when the club spent lavishly on players, very often employing former North American Soccer League players, and even U.S. National Team members. “This team is strictly amateur,” said Barone, who takes the club into Harrisburg for a Tuesday first round game against the USL2 City Islanders . Barone will be without two of his top players, midfielder Darwin Iriarte and defender Jean Pierre Ahoua, who could not get off work to make the trip. That is not to say Barone does not expect to do well in the game. “We’re an experienced team,” he said. “There’s a lot of flair in the team. We could surprise them,” he said of the City Islanders, who have several players on loan from MLS’ Philadelphia Union, including former Red Bull Nick Zimmerman. Another former Red Bull, Danny Cepero, plays in goal for the City Islanders. While the Brooklyn Italians club has been around since 1949, they are in the first year of fielding a team in the National Premier Soccer League, after going more than a decade without a men’s team. “We are putting all our resources into the youth clubs,” Barone said. Still, Barone says he is fielding an experienced team that includes pro hopeful Semso Nikocevic, who had trials both with Real Salt Lake and the Rochester Rhinos. The Rhinos wanted the midfielder, but did not offer him a reasonable salary, according to Barone. Former Clemson player Alan O’Hara anchors the team’s defense. The Italians are currently 5-1 in the Northeast Division, in a neck-and-neck race with the Red Bulls Academy for first place. If the Italians advance, they have two possibilities for the second round. They will either face the Long Island Rough Riders at Cy Donnelly Stadium in South Huntington, N.Y., or their old rivals the New York Pancyprian Freedoms at a site TBA.

Lamar Neagle, Mike Zaher and player-assistant coach Ian Fuller scored for the Battery in its 3-1 shootout win after the game ended scoreless after 120 minutes. The Fire missed two and had a third attempt saved by goalkeeper Tim Melia. Struggling D-2 Pro League team AC St. Louis held MLS 's top team, the Los Angeles Galaxy, for a half before falling, 2-0, on second-half goals by Chris Klein and young Brazilian Juninho, his first for the Galaxy. Chivas USA edged D-2 Pro League leader Austin, 1-0, on Jesus Padilla's goal in Fullerton, Calif. Steven Lenhart's goal in stoppage time gave the Columbus Crew a 2-1 win over the Rochester Rhinos. As of this morning, there may be 10 matches with video and/or audio. Several teams caught a lot of flack for not doing more to get the games accessible. I was one who jabbed at many of them. Just stupid they don't stream these matches, USSF is to blame -- then the clubs.

I originally posted this in the LHUOSC section, based on a podcast with the league honchos, but have found no confirmation. It is not in the LHUSOC Open Division Handbook. It does make sense, though, in that the league works similarly to the PDL - NPSL, as a summer-only college-player-based league.

MINNEAPOLIS — Minneapolis City SC has been disqualified from the U.S. Open Cup after the club found itself in conflict with current cup policy. Per the U.S. Soccer Federation Policy manual, Open Division teams forfeit their eligibility upon switching leagues after the Open Division entry deadline has passed. Minneapolis City was awarded an Open Division berth ahead of qualifying for the 2017 Open Cup, then defeated Oakland County FC of Rochester Hills, Mich. 2-1 in extra time in the second qualifying round on Oct. 22, 2016. On Dec. 14, City officially left the Premier League of America (PLA) to join the NPSL, citing travel costs and a desire to establish local rivalries as its motivation. City was alerted to the then-potential eligibility conflict by former PLA leaguemate RWB Adria of Chicago.
