In no particular order:
Tommy Tiernan (hilarious... the only person on the Just For Laughs Tour to be asked to do a full set 2 years running)
Zach Galifaniakis
Mitch Hedberg
Bill Bailey
Jack Dee (unbelievalbe that I haven't seen him mentioned yet!)
Dennis Leary & Bill Hicks (they're pretty damn similar, tbh)
Also, I saw this one guy on the just For Laughs festival once. He was American, had quite whiney voice (quite like Ray Romano), and he didn't face the audience directly... instead he stood at a kind of diagnol angle to them, looking sideways. He also had his hand to his face nearly the whole time, and told some strange anecdotal type jokes. If anybody can remember his name, I'll owe you big. The guy was hilarious.
Jeremy Hotts is the guy you are looking for.

Ah, nice one... rep to you, good sir! Turns out it's actually Jeremy 'Hotz' and he is Canadian, I always thought he was a New Yorker for some reason. I just find his self-depricating humour hilarious. For those who may not be as familiar with him...

That spelling error is a little embaressing since I acctually went to see him on the last Just for Laughs tour that came to town, basically just for him and Russell Peters. If I liked someone enough to spend money on him you would think that I would remember how to spell his name! :D

Dave Attell is one of my favorites. Ditto for Lewis Black.
In fact, my wife and I saw the Comedy Central tour they did a few years back which featured Mitch Hedberg too. That was probably Mitch's last tour.
I saw that show at the Rosemont Theater in Chicago a few years ago. It was frickin brilliant.

One Australian addition here -- Mick Molloy.
Mick's got a dry, sarcastic sense of humor that just bites away at the inner reaches of one's soul, and isn't afraid to say what he thinks, no matter how controversial it might be, as long as he's funny.
In addition to showing off his comic acting range in a couple of recent Australian flicks such as "Crackerjack" and "Bad Eggs" (and two movies on DVD worth making overseas purchases, mind you, they're that good), he can be heard most weekday afternoons from 12 noon Melbourne time on Triple M radio here.
One sample from his "Tough Love" (http://www.mmmelb.com.au/shows/mick/index.php) show, from some months back, about Paris Hilton's dog (and I'll use the "result" tags in the event that this could offend)...
"Paris Hilton's dog is pregnant. Yeah, I know. I saw the video.
"Amazingly, there was less 'doggy-style' in that video than in Paris' video."
Funny stuff. :D

Absolutely! One must get a copy/copies of "Crackerjack"/"Bad Eggs" -- even if you have to go to Amazon.com to do it.

Out of interest -- which flick? Or both?
Lots of quotable lines in each one.
And in that spirit, Bob Franklin and Judith Lucy are quite funny as well.

Another guy I only caught for the first time on the Thanksgiving episode of Conan was Demetri Martin. I thought he was pretty good, so I d/led his Comedy Central act (we don't get Comedy Central here, so I don't consider it wrong). While the final song bit was a bit weak, the rest was pretty hilarious, and quite like a mix of Mitch Hedberg & Zach Galifaniakis in a good few ways. He needs to iron his stuff out a bit, but he looks very young, so I doubt that will be much of a problem.

Steve Martin for Let's Get Small alone
Mitch Hedberg, may he rest in peace.
Dave Attell ("We call ourselves the Un********ables"
Bobcat Goldthwait (once you get past the screeching, he's pretty brilliant)
Eddie Izzard
Bill Cosby
Johnathan Winters
and for some reason I find Monique Marvez to be hilarious, although I'm not exactly sure why that is. Maybe I just have a thing for plus size Latinos. Who knows? ;)
