Does getting relegated to a lower division, or getting promoted upward have a effect on teams and their rivalries. Is there a club that considers a lower division team its rival, or do the team and its fans form new rivalries when promotion or relegation happens?

Burnley and Blackburn have played each other for only two seasons in the last quarter of a century, but they still consider each other their bitterest rival.

Yep. Same for Wednesday-United and when they were done, Southampton-Pompey.

In the most part rivalries are local so you'd hate them even if they played in the conference.

Nope, relagation doesn't have any effect on the chippiest local derby, just ask Bristol City and Bristol Rovers

pompey vs southampton 2004 - same league - 54 imprisoned so far - over 300 involved in a large scale riot
pompey - southampton 1994 (i think) - Alan knights testimonial (a 'friendly') at least 30 arrests and a large scale riot - (less caught than this years game as it's all filmed these days) - different leagues
don't think it matters what league and absence making the heart grow fonder is a myth!

So true. I've been to see Blackburn a few times and one of the few things they ever sing about is Burnley.

Leeds has rivalries with anyone in the Yorkshire, with Sheffield United being the strongest rivalry.
